摘要

It is well-known that DNA-protein complexes such as that between lac operator and repressor can be trapped on nitrocellulose filters. The complex formed in the dark between DNA-photolyase and irradiated IP T7 phage DNA can be trapped the same way. If the complex is photoreactivated, the labeled DNA passes through the filter. Based on these observations, a new and rapid assay has been devised to measure photolyase. The assay permits the molar concentration and the percent purity of the enzyme to be calculated and it is this parameter, rather than the usual specific activity which is used to follow enzyme purification.
