We have established conditions which allow the in vitro growth and cloning of human T-lymphocytes, as well as methods for selection and quantification of 6-thioguanine resistant (TG/sup GAMMA/ mutants. In particular, we have established a procedure to measure the in vitro induction of TG/sup GAMMA/ mutants by gamma irradiation having defined the phenotypic expression time and optimal selection conditions. 24 figs., 6 tabs.
