Establishing the Order of Human Chromosome-Specific DNA Fragments

While the coupling of the cosmid and large fragment approaches offers a significant improvement over either method alone, the establishment of a single contig for chromosome 19 is not a certainty. The inability to clone some DNA sequences, the non-uniform representation of cloned sequences, and the presence of highly repeated DNA sequences, which may confuse the overlap analysis, all confound this ultimate goal. It is likely that new methods may simplify or replace some of the steps involved and research into and development of such methods is critical to any large scale commitment to ordering and subsequently understanding the human genome.
