In this report, we investigate the role of ECM proteins and growth factors inanchorage-independent survival and growth of a murine mammary carcinoma-derived cell line, designated SP1. We demonstrate that SP1 cells adhere well to FN and VN and express a number of integrins, including a5beta1, alpha v beta 3, alpha 6 beta 1, alpha 3 beta 1, and alpha 2 beta 1, the latter being upregulated under anchorage-independent conditions. We also demonstrated by partially removing FN from 7% FBS or adding FN to 1% FBS that FN is required for SP1 cell colony growth.
