This is a Standard Operating Procedure (SOP) to glycerolize and deglycerolize human red cells in the 800 ml primary polyvinylchloride (PVC) plastic collection bag system. The glycerolized red cells are concentrated and the supernatant glycerol removed prior to freezing. The glycerolized red cells are washed in the Haemonetics Blood Processor 115. Highlights of this revised SOP include a sterile docking procedure for glycerolizing the red blood cells, a change in centrifugation speeds, and new procedures for thawing the red blood cells prior to deglycerolization. Freezing in the primary PVC collection bag reduces the potential for contamination, eliminates the need for a special freezing bag, reduces the volume of wash solution, permits biochemical modification of indated and outdated red cells, increases the storage capability of the -80 C mechanical freezer, and reduces the cost of freezing a unit of red cells by 25%.
