The effects of retinoic acid (RA) in breast cancer have been widely studied. Treatment with RA leads to either growth arrest or apoptosis in breast cancer cells. The cyclin-dependent kinase inhibitor p2l WAF1/CIP1 may be involved in terminal differentiation associated growth arrest. We showed in this study that PML/RAR alpha increased the transcription of p21WAF1//CIP1 gene and the activation was further induced by RA treatment. Deletion analysis revealed a region upstream of the p2lWAFJ/ClPl promoter that is required for transactivation by PML/RAR alpha. Transient transfection of PML/RAR alpha in cells increased the endogenous p2l WAF1/CIPl protein levels.
