摘要

The ultimate goal of the project described in the present report is to perform in vivo screening experiments of phage-displayed random peptides libraries (RPLs) in human breast cancer patients, in order to identify peptides that will home specifically to their tumor tissue. In future work, these breast tumor-homing peptides will be used to develop novel breast cancer therapeutics. The work described in this annual report includes: (1) Construction of five new RPLs. (2) Toxicity testing of intravenous injection of peptide-phage in animals, which is detailed in an enclosed IND application. The ND needs only minor additions for approval by the FDA. (3) Several in vivo screening experiments in mice with tumors. The in vivo screening experiments we have performed in mice, with essentially the same protocol proposed for human screening, as well as other toxicity testing experiments, provide evidence that in vivo screening with phage-displayed RPLs is safe.
