The uptake of D-aspartate by rat cerebral cortex slices was found to consist of two processes: an energy-requiring system, and possibly nonspecific binding. The accumulation of D-aspartate was inhibited by .0002 M concentrations of hydrazine, monomethylhydrazine (MMH), and 1,1-dimethylhydrazine (UDMH), but not by 1,2-dimethylhydrazine (SDMH). At a higher concentration (.005), hydrazine inhibited uptake by 50%, while UDMH produced a 25% inhibition. Uptake of D-aspartate in cerebral cortex slices from rats given 1.6 mmoles/kg. UDMH (LD50) and sacrificed before any convulsions was no different from control values. The accumulation of D-aspartate in cerebral cortex slices from rats given the same dose of UDMH but allowed to convulse was greater than that of the controls. The lack of inhibition of uptake in cerebral cortex slices from UDMH-injected animals militates against the suggestion that UDMH might produce convulsions by interfering with active transport across neuronal membranes.
