Chromatographic Isolation of Some Cellulose Oligosaccharides

The preparation of cellotriose and cellotetraose from cellulose is described. The compounds were obtained as the pure alpha acetates by column chromatographic separation of the methanol soluble portion of a partial acetolyzate of cellulose. Application of 15 grams of the acetolyzate to a 6.5- by 50-centimeter silica gel column, followed by elution with benzene-ethyl acetate (1:1) gave 2.1 grams of alpha cellotriose hendecaacetate and 2.0 grams of alpha cellotetraose tetradecaacetate. Elution from a free-flowing column required 12 liters of irrigant and took about 4 days. Chromatographically pure crystalline cellotriose and cellotetraose were obtained by standard deacetylation procedures. (Author)
