Isoimmune Responses in the Rhesus Monkey: Hemagglutination, Leukoagglutination, and Leukotoxicity

Normal, mature rhesus monkeys were isoimmunized with fresh, viable leukocyte preparations from single donors. Treatment consisted of injections into 1 intrapalmar and 9 intracutaneous sites, at 5- to 7-day intervals, for a total of 20 treatments. Resultant isoantisera showed strong and repeatable hemagglutinating, leukoagglutinating, and leukotoxic activity in in vitro test-systems. Results of tests against erythrocyte and leukocyte panels indicated isoantigens peculiar to cell type as well as those shared in common. It was also evident that each test-system was detecting different isoantigens despite some duplication. No normal isohemagglutinins were detected, although widely reactive hemagglutinins were demonstrable in the serum from females post partum. Isoimmume hemagglutinins reached titers of 1:512, retaining low-titered activity for 90 days after treatment. Hemagglutinating activity was associated with 60% of definitely positive leukoagglutination and leukotoxic reactions indicating the possible existence of a histocompatibility hemagglutinogen system. Serologic procedures developed or modified for use with the rhesus monkey have been described in detail. (Author)
