Neuronal Changes Induced by Intrathecal Vincristine Sulfate.

Intrathecal administration of vincristine sulfate in a child with acute lymphocytic leukemia produced neuronal changes characterized by clumped Nissl substance separated by clear areas and acidophilic rhombohedral crystals. Similar changes were induced experimentally in rabbits. The clumped Nissl substance corresponded to conglomerates of free ribosomes admixed with short fragments of granular endoplasmic reticulum. The clear areas were argentophilic and consisted of skeins of interwoven neurofilaments. The crystals appeared as either parallel rows of punctate osmophilic densities or arrays of circular profiles, depending upon the plane of section. The possible derivation of the crystals from the neurofilaments is discussed. (Author)
