Different Mechanisms of Drug-Induced Release of Noradrenaline and Its Congeners Alpha-Methylnoradrenaline and Metaraminol

Tritium-labelled noradrenaline, alpha methylnoradrenaline or metaraminol was given i.v. to mice. All three amines were efficiently taken up by the heart. Reserpine, amphetamine, protriptyline, and nialamide were shown to have amine-releasing properties, though with differential effects on the three amines, resulting in characteristic 'release profiles'. The interaction between the releasing agents was also investigated. The results give an insight into the various mechanisms involved in amine uptake, storage, release and degradation by adrenergic nerve fibres and into the mode of action of the drugs investigated.
