Biologic Reactions of Cellular Antibodies with Special Reference to Their Immuno-Pathological and Immuno-Chemical Properties

The omentum cells, cultivated from the omentum pieces of bovine serum albumin (BSA)-sensitized rabbits and grown in a special culture medium, were found to contain anti-BSA antibodies of 19S and 7S classes. From the cell extracts, the gamma-globulin fraction containing the antibodies was prepared with sodium sulfate at 18 per cent saturation. From the protein preparations, the antibodies were fractionated by gel filtration with Sephadex G-200 or by sucrose density gradient. Antibody titers were measured by an indirect hemagglutination test or a quantitative precipitin test. The antibody of 19S class was particularly noteworthy in the biological property. The antibody had a strong ability to induce a passive cutaneous anaphylaxis in a homologous animal (rabbit) but not in a heterologous animal (guinea pig). Such an biological activity was abolished by treatment with 2-mercaptoethanol, in a parallel with the decreased antibody titers. The biological activity of the 19S class antibody was also characterized by its rapid sensitization of rabbit skin and its short persistence in the skin site. On the other hand, the 7S class antibody induced a passive cutaneous anaphylaxis in a heterologous animal but not in a homologous animal, indicating an biological action different from that of the 19S class antibody. The treatment with 2-mercaptoethanol did not influence the 7S class antibody. The biological significance of these antibodies in an allergic tissue response was discussed. (Author)
