A high yield of the purified K antigen was obtained when cells of Vibrio parahaemolyticus A55 were boiled in 3% NaCl for the liberation of K antigen and the clarified supernatant was subjected to zone electrophoresis to elute the hexosamine peak fraction Average yield: 30mg of the purified K antigen per 4g dry weight of cells. The antigen obtained showed a high degree of serological activity and specificity. On centrifugal and electrophoretic analyses it was shown to be homogeneous. On its degradation study, a maximal liberation of hexosamine (ca. 40% calculated as glucosamine) was obtained by heating it with 10 N NCl at 100C for 1 hr. Paper electrophoresis of the hydrolysate separated three components reacting with silver nitrate and ninhydrin reagent, which travelled toward the cathode at pH 8.9 and toward the anode at pH 11.6. On the basis of these facts, the major components of the antigen were presumed to be acidic aminosugars. Their identity, however, was not established. (Author)
