The relationship between dephosphorylation and recovery of inhibited brain acetylcholinesterase (AChE) following intravenous administration of 32P-isopropyl methylphosphonofluoridate (Sarin) to rats was studied. Some decrease in inhibition of brain AChE activity occurred between 1/2 hour and 48 hours after poisoning, concurrent with comparable dephosphorylation of the enzyme. Injection of 44 mg/kg of monoisonitrosoacetone resulted in a further decrease in inhibition of AChE activity over and above that occurring spontaneously, accompanied by additional dephosphorylation of the enzyme. Loss of phosphorus in vivo appeared to take place exclusively as isopropyl methylphosphonic acid. No significant removal of phosphorus bound to AChE as methylphosphonate was apparent under these conditions. (Author)
