Electron Microscope Studies on Several Phosphatase Activities in Neurons and Gliacytes Infected with Japanese Encephalitis Virus

Ultrastructural and microchemical analyses were made on the multiplication of Japanese encephalitus virus (JEV) in mouse hippocampus neurons infected intracerebrally or intraperitoneally with JEV. The viral factory appears in the cytoplasm of infected neurons at least 3 days after intracerebral or intraperitoneal inoculation of JEV, which consists of a large amount of unknown dense masses associated with clusters of fine vesicles, numerous free ribosomes and virus particles in various development stages. The mature JEV consists of an electrondense core and a less dense envelope, and the former has been revealed to be an ATPase active site. The premature virus is found within the smooth-surfaced cisternae. It consists of a dense core and a less dense surrounding area, but two parts are variable in sizes and shapes, and their contour is ill-defined, showing ATPase final reaction deposits. Many mitochondria are swollen and deformed to a ring-, racket-, or horseshoe-shaped feature. In the electron-lucent area of the swollen mitochondria, numerous dense particles 140 to 260 A in diameter and a few coiled filaments 70 A in width have been onserved, suggesting that such particulate elements may be precursor of JEV, since they are composed of nucleic acid. The presence of zinc elements in mouse brain is confined in the hippocampus neuron. Their localization has been analysed to be lysosomes containing dense granules. While, the JEV-infected neurons are devoid of zinc elements. (Author)
