The purpose of this research is to eradicate chemotherapeutically malarias in the human host; drug resistant strains of P. falciparum constitute the primary target. The aim is to suggest molecules for synthesis and testing which have a high probability of success as effective antimalarials. Results include numerous Hansch analyses, Free-Wilson analyses, and molecular orbital calculations on known or potential antimalarials. Data reported by Elslager and Stogryn were subjected to quantitative structure-activity models. Free-Wilson analyses were applied to antimalarial data on phenanthrene methanols, triazines, and quinazoline-pteridines. Several compounds are suggested which have high probability of success as antimalarials. The interpretation of the mode of antimalarial activity for chloroquine analogs was investigated via molecular orbital calculations and mathematical models. The mechanism of DNA intercalation for these analogs is discussed. (Author)
