Specificity of a Rabbit Antihuman Lymphocyte Serum

The specificity of antibodies produced after prolonged immunization of rabbits with purified human lymphocytes from a single individual (B.L.) was examined. Lymphocytes and leukocyte-platelet packs from B.L. and from ten other individuals unrelated to B.L. and to each other were used. The antiserum had a cytotoxicity titer of 1:256 against B.L. lympnocytes and against lymphocytes from eight other individuals. A titer of 1:128 was obtained against lymphocytes from the other two individuals. Absorption studies at 1:64 and 1:4 dilution of the antiserum indicated that separate absorptions with purified lymphocytes (1:64) or leukocyte-platelet packs (1:4) from each of the ten other individuals, as well as from B.L., completely removed cytotoxicity against both the absorbing lymphocytes and B.L. lymphocytes (with one exception at the 1:4 dilution). Absorption studies at the 1:64 dilution indicated that residual cytotoxicity existed for B.L. lymphocytes at the point where cytotoxicity was just removed against the absorbing lymphocytes from each of the ten other individuals.
