Phosphonofluoridate Inhibition of Acetylcholinesterase:An Exceptionally Rapid Reactant.

Both stereoisomers of (3,6-dioxa(8-(2,4-dinitrophenoxy) octyl) methylphosphonofluoridate (I) react rapidly with eel acetylcholinesterase. Reaction rates are reported. Despite the very high rate of reaction with AChE,the toxicity of I is only one-ninth that of Sarin. Enzyme inhibited by I is resistant to reactivation by TMB-4. Implications in prophylaxis and therapy of G-type agent poisoning are discussed. (Modified author abstract)
