Hypersensitivity to 'Schistosoma Mansoni' Antigens. I. Immunochemical and Biological Characterization of an Antigenic Extract.

An adult Schistosoma mansoni extract (FT 1) was obtained in good yield (1000mg per 100,000 worms) after a single freezing and thawing cycle. This fraction was highly reactive by both Prausnitz-Kustner (P-K) and passive cutaneous anaphylaxis (PCA) tests with sera from schistosome infected rats. Adult, cercarial and egg antigens were also tested by the P-K assay. In general, adult antigens were more reactive than cercarial and soluble egg antigens. Rats produced detectable levels of reaginic antibody to FT 1 with 4 weeks after initial infection and showed a clear anamnestic response after reinfection. The ability of various antigen preparations to induce reaginic antibodies in rats was also studied. The FT 1 preparation was found to be composed of 10 per cent carbohydrate, 90 per cent protein and a trace of ribonucleic acid. The fraction contained materials with a broad range of electrophoretic mobilities as shown by both disc gel electrophoresis and immunoelectrophoresis. (Author)
