Replication of Japanese Encephalitis Virus.

Preliminary studies of the structure of Japanese encephalitis virion RNA indicated that the 5'-terminus had a 7-methyl-guanine cap and that the 3'-terminus had little or no polyadenylic acid. Definitive structural studies have not been completed due to difficulties in obtaining sufficient material. Two-dimensional electrophoretic maps of RNase T1-resistant oligonucleotides derived from virion RNA propagated in vertebrate and invertebrate cells were identical. Attempts were made to determine th 3'-terminal base sequence of virion RNA by end-labeling followed by base-specific chemical cleavage. The RNA accepted label poorly by comparison to RNAs from other sources, suggesting the presence of a unique 3'-terminal structure.
