Research and Development on the Modification of Stroma-Free Hemoglobin for Use as a Blood Substitute

Chemical modification of stroma-free hemoglobin (SFH) using a series of ribose phosphates has been achieved. The P50 is 50-70% improved and the intravascular retention time increased by two to eight times (species dependent). The dial modified SFH has normal oxygen carrying capacity and maximally altered energetics. It is stable at 4 C liquid storage and maximum met-Hb is 4.5%. Details of the reactions indicate a significant yield 90% can be obtained by iteration of the reaction steps. Extensive purification is not needed.
