Stability Studies of Immobilized Cholinesterase and Parathion Antibodies

The enzyme, acetylcholinesterase EC 3.1.1.7, was immobilized by seven physical and chemical methods and then assayed using a simple, but accurate electrochemical three electrode system. The long term stability of these immobilizations was studied under various storage conditions. The effects of some possible environmental contaminants on the relative cholinesterase activity was studied. It was found that the triazine and collagen methods are the most promising ones for this enzyme under varying conditions of storage and use. Parathion antibodies were bound on piezoelectric crystals, using both the chemical cross-linking (glutaraldehyde/BSA method), and physical adsorption methods. The chemical approach gave rise to longer stability of over 2 months.
