Human brucellosis, a major public health problem in most developing countries, is frequently diagnosed by antibody detection, because attempts to isolate the bacteria from blood are often unsuccessful. Agglutination, the routine serological assay, is insensitive and difficult to interpret reproducibly. RIA and enzyme immunoassay are helpful for detecting anti-brucella antibody classes. We have developed a 'Dot-ELISA,' in which whole bacteria (whole-cell antigen, WCA, lipopolysaccharide (LPS), and soluble sonicated antigen (SSA) are used to detect IgG, IgM, and IgA antibodies to brucella. We studied 47 sera from 32 culture-positive and 15 seropositive patients (by ELISA) with Brucella melitensis infection. Reprints. (AW)
