Evaluation of Membrane Systems for Washing/Deglycerolizing Packed Red Blood Cells (Annual Report, February 10, 1987-February 9, 1988).

The objective of this contract is to develop a membrane-based process for deglycerolizing previously frozen, packed red blood cells (RBCs) prior to transfusion. This membrane-based process must reduce the glycerol concentration in one unit of RBC solution to less than 1 wt% using about 2000 ml of wash solution in 35 to 45 minutes. The plasma hemoglobin concentration in the washed RBCs must be less than 150 mg/dl. During the second year of the contract the performance of our membrane-based deglycerolization process exceeded the original performance targets. We also demonstrated that at least eight units of RBCs can be effectively deglycerolized using the same membrane module. If necessary, the modules can be cleaned for the reuse using a procedure we developed. Our optimization studies showed that the efficiency of deglycerolization can be improved by concentrating the RBCs before washing and by promoting better mixing of the RBCs and the wash solution within the blood bag.
