Fibronectin (FN) has been detected on red blood cell (RBC) surfaces using fluorescent microscopy and transmission electron microscopy following the transfusion of incompatible blood to dogs. This study was an attempt to demonstrate and quantitate RBC bound FN by immunofluorescent flow cytometric techniques following a hemolytic transfusion reaction (HTR) in dogs. Collateral testing was performed for other known opsonins, ie. immunoglobulin G (IgG) and the C3 component of complement (C3). Red blood cell bound FN was not detected using this study's techniques; RBC bound IgG and C3 were detected. Previous findings of RBC bound FN after an incompatible blood transfusion reported low numbers of FN molecules per cell when compared to amounts of RBC bound opsonins IgG and C3. The sensitivity of flow cytometry may be insufficient to detect RBC bound FN using the labelling methods and fluorescent antisera in this study. Keywords: Blood chemistry; Blood analysis. (kt)
