Optimal Blood Sampling Times for Diazepam Pharmacokinetics in Rhesus Monkeys

Diazepam (0.2/mg/kg) was intramuscularly administered to a rhesus macaque in the right hind limb. Blood (3 ml) was collected via a left saphenous catheter. Serum was analyzed for diazepam concentrations by gas chromatography-mass spectrometry employing selective ion monitoring. Maximum serum concentration was 128 ng/ml at 34 min. Optimal blood sampling times appear to be 0,5,10,15,25,40,60,90,120, and 240. The absorptive phase for the monkey was similar to that of man but the monkey's elimination phase was much quicker. (AW)
