This final report describes our research effort in the generation of high-binding-affinity monoclonal antibodies specific for soman (GD). The different parameters that were studied included the use of novel GD conjugates and analogs, GD stereoisomers, in vivo boosting of immune mice with sublethal doses of free soman, in vitro stimulation of GD-immune mouse spleen cells with GD, and the generation of human monoclonal anti-GD antibodies. We were successful in isolating a monoclonal antibody specific for GD with an approximate IC50 value of 7 x 10-8M, which is significantly higher than previously generated anti-GD antibodies. This antibody, termed GD1 (also called GD2H which is a subclone of GD1), was shown to increase the in vivo LD50 value of soman by three to four fold in BALB/c mice treated with GD1 or GD2H.
