This research demonstrated the feasibility of using primary mixed liver cells ina bioreactor for the production of a pure population of immunogenic vesicles. This work has shown that mechanically disaggregated cells from adult CD rats are capable of maintaining their viability, morphology and biochemical phenotype for prolonged periods of time (more than 19 days) without the addition of serum, mixtures of adhesions, cort-costeroids, or dimethyl sulfoxide. A significant innovation for this work was the development of a microporous, gelatin-nylon cell carrier. When liver cells were encapsulated in sheets of the gelatin sponge, they aggregated into clusters with a morphology reminiscent of whole liver tissue. (JES)
