The unsymmetrically substituted trifluoromethylphosphorane 1 was designed as ahapten to generate the catalytic antibody of phosphoryl transfer activity. Phosphorane 1', the rigid analog of phosphorane I was first synthesized and was stable for at least 12 hours in aqueous conditions over pH range of 1 to 11. The targeted phosphorane 1 was successfully synthesized but was rapidly decomposed in the hydrolytic stability test. It is postulated that the flexible S-S pyridyl arm somehow intereacted with the phosphorus center and made it unstable. The effort of replacing it with other stable linker arms in order to enhance its aqueous stability is currently being made.
