In the present study, it was demonstrated that the immunostaining patterns of thebullous pemphigoid antigens BP230 and BP180 as well as the cytoskeletal intermediate filament keratin-5 were altered after exposure of human keratinocytes to HD. The concentrations of HD and the post-incubation times after which the effects on the BP antigens and keratin-5 were observed, correlated fairly well with the in vivo data on the onset of blister formation in human skin. It was suggested that the reduced capacity of BPl8O, BP23O and keratin-5 to react with their specific antibodies might result from degradation by certain proteases. Furthermore, it was demonstrated that exposure of human keratinocytes to HD did not disturb Ca2+ homeostasis. On the other hand, receptor-activated Ca2+ signaling in RD-treated human keratinocytes was obviously inhibited. Based on the present findings, application of protease inhibitors might provide a real possibility to prevent blister formation in victims of RD exposure.
