For mass multiplication and sustainable utilization of medicinally promising herb Ocimum tenuiflorum, micropropagation protocol has been developed. In this study, multiple shoots were initiated more from nodal explants in MS medium supplemented with various concentrations of BAP and Rn. The synergistic effect of BAP (1.0mg/l) induced twelve adventitious shoot buds from the nodal explants. The frequency of shoot organogenesis was highest (100%) in nodal explants treated with 1.0 mg/1 of BAP. The meannumber of shoots observed in the combination of BAP (1.0mg/l) with IAA (1.0mg/l) was six from nodal explants. Among the various growth regulators tested for rooting, IAA at 4.0 mg/1 showed best results, where five roots were initiated after 15 days of culture. The developed plantlets were transferred to small pots containing soil, cow dung compost and cocopeat (2:1:1 ratio) and maintained in the green house for hardening. This investigation definitely serves as model system for mass multiplication and commercial utilization of other medicinal plants.
