A successful protocol has been developed for rapid clonal propagation of Aegle marmelos using nodal segments. The explants were cultured on MS medium augmented with different concentrations (0.5-5 mg/1) of BA, KIN and 2ip and combinations of BA (0.5 mg/1) with KIN/2ip (0.5-5 mg/1) and KIN (2 mg/1) with BA/2ip (0.5-5 mg/1). 0.5 mg/1 BA induced the maximum number (8.40) of shoots. In vitro raised shoots were rooted on half strength MS medium supplemented with three auxins, NAA, IAA and IBA (0-2.5 mg/1). IBA (2.5 mg/1) was found to be better than IAA and NAA. The regenerated plantlets were acclimatized with 65% survival rate. Inter Simple Sequence Repeat (ISSR) marker assay was employed to validate the genetic fidelity of these regenerated plantlets. All banding profiles from micropropagated plants were monomorphic and comparable to those of the mother plant, thus ascertaining the true nature of the in vitro raised plants.
