Collection, handling, fixation, histological and storage procedures for taxonomic studies of terrestrial flatworms (Tricladida: Terricola)

Methods for the collection, handling, fixation, histological, and storage procedures used in the taxonomic studies of terrestrial flatworms (land planarians) are provided. In general, specimens are best narcotised in 10% ethanol, and morphology preserved using a formaldehyde calcium cobalt fixative; long-term specimen storage is in 80% ethanol. Specimens are cleared for examination in terpineol, then processed to paraffin wax, and serially sectioned. Sections are stained using the AZAN or MSB polychrome methods. Specimens for DNA investigations are fixed in 100% ethanol.
