A Direct Introduction of the Mercaptoacetic Acid Unit into Amino Acid Esters

The reaction of amino acid esters with l,3-oxathiolan-5-one la provided thiol amides 3.(alpha)-Mercapto amides are useful subunits in metal ion complexing agents.1 Acylation of amines with activated mercaptoacetic acids in which the sulfur is protected as a trityl or TBS group, followed by deprotection, affords effective complexing agents for technetium and other metals. Although this multistep route has been often used to prepare individual compounds, it is less desirable for the production of a combinatorial array.
