4-Hydroxycyclohex-2-en-l-one and its derivatives have been used as the key chiral building blocks in a number of natural product and complex molecule syntheses. This review describes the many and varied methods for preparing the title compound in an enantioselective manner and also details how it has been utilised in natural product synthesis.The wide-ranging structural diversity and varying biological activity profiles of the molecules in Figure 2 demonstrates how useful enone 1 is as a synthetic building block. Given the desire to produce single enantiomers of such natural products, several methods for producing enantiomerically enriched enone 1 have been disclosed and will be discussed herein.
