Electrophoresis is commonly used in genetic studies and for line identification in cultivar improvement programs of many crops including soybean. Both starch gel electrophoresis and polyacrylamide/starch gel electrophoresis have been used in earlierstudies. While these systems have worked successfully for most enzymes, resolution of alcohol dehydrogenase (ADH EC 1.2.3.4) from soybean has proved problematic. Cardy and Beversdorf (1984) used starch gel electrophoresis and reported that ADH, though present, was poorly and inconsistently resolved. Rennie et al., (1989) indicated that whereas all variants of ADH could be separated on the polyacrylamide/starch system used by Bult et al., (1989), results were inconsistent with the starch gel system.
