The most popular use of DNA biosensors is DNA profiling. Also referred to as the genetic fingerprinting, this method allows DNA molecules be sequentially separated by size and then detected via amperometric sensing. Despite numerous progress made on this subject, however, there are some outstanding issues related to resolution and signal detection. In this article, a Microchip Capillary Electrophoresis (MCE) was designed and fabricated for studying the effect of sample injection strategy on signal detection. The biochip was tested using different DNA samples from which it was discovered that injecting a sample through TAE buffer significantly improved the signal resolution, resulting in unique electropherogram for separation voltage as low as 12 V/cm.
