The effect of 2, 4-D on callus induction and regeneration in three popular varieties viz., Mahsuri, Kranti and IR 54 was studied. It was observed that higher the concentration of 2, 4-D the higher is the callus induction percentage but lower is the regeneration percentage. The somaclones obtained from the three cultivars were grown in field and selfed seeds were grown for next generation for somaclonal variation. Plant height increased in 40 and 37.5% somaclonal plants of Mahsuri and IR 54, respectively. Number of tillers and economic yield also increased in Mahsuri and IR 54. However, only a line from IR 54 (line no.8) was found promising from agronomic point of view. It has increased economic yield and shorter plant height. with respect to parent.
