The phosphatidylinositol 3-kinase (PI3K) pathway regulates cell growth, survival and proliferation in response to many stimuli and is frequently overactive in cancer cells.' The p1110 catalytic subunit of PI3K is a lipid kinase that generates phosphatidylinositol 3,4,5-trisphosphate (PI3,4,5P_3), a product dephosphorylated by PTEN. The PI3K product PI3,4,5P_3 is a key second messenger that relocalizes and induces the activation of PDK1 and Akt, important signaling kinases with many downstream targets involved in mediating cellular responses.
