The level of expression of the preproorexin gene was studied in rat hypothalamus cells 2, 4, and 6 h after administration of lipopolysaccharide (LPS) at doses of 25 and 500 μg/kg. The level of gene expression in rat hypothalamus cells 2 h after administration of both small and large doses of LPS increased, which is indirect evidence for an increase in orexin synthesis in neurons. No changes were seen at 4 and 6 h. Comparative analysis of the results of previous immunohistochemical studies and the data presented here allow the mechanisms of the reactions of orexin-containing neurons to different doses of LPS to be assessed: there was a predominance of orexin utilization processes over synthesis processes in hypothalamus cells after administration of subseptic (500 μg/kg) doses of LPS.
