The effects of the selective N-methyl-D-aspartate (NMDA) glutamate receptor blocker Ro 25-6981, given at different time points at a dose stimulating neurogenesis, on the formation of a new skill in adult rats with different learning abilities were studied. Male Wistar rats were trained for five days to find a hidden platform in a Morris water maze. The animals' ability to form a spatial skill was assessed in terms of the time taken to reach the platform. All rats receiving Ro 25-6891 13 days before retraining (with the contextual stimuli and platform in different positions) demonstrated impairment of skill formation. Ro 25-6981 given 29 days before retraining selectively facilitated formation of the skill in animals with initially low learning ability.
