Purpose: Titanium is a bio-material of implant and it is important to understand its coating structure that bonds to the host bones. We investigated cellular responses of the coating of titanium with calcium phosphate (CP)/Collagen I (COL-I) complex in vitro. Methods: To investigate the bone-integrating ability of CP/COL-I complex, we set up four experimental groups: titanium with no coating (control), with CP coating, with multi-porous CP/COL-1 coating, and with dense CP/COL-I coating. MC3T3-E1 cells were cultured on the coatings, and cell morphology was observed by scanning electron microscope (SEM), cell proliferation measured by the MTS assay, and the gene expression of alkaline phosphatase (ALP), COL-1, and osteoblast (OC) examined by reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis. Results: MC3T3-E1 cells grew significantly faster by roughly 20% in CP/COL-I coating, while no increase was seen in CP or no coating. Significantly higher gene expression of COL-I, ALP, and OC was observed in multiporous and dense CP/COL-I coatings than CP-only coating or no coating (P < 0.05), and there was approximately a 30% increase in dense CP/COL-I coating compared with multi-porous CP/COL-I coating. Conclusions: The titanium coated with CP/COL-I exhibits better cellular response in vitro, which may be due to the activation of bone matrix proteins genes stimulated synergistically by CP/COL-I coating. The concentration of COL-I in the coating may also be a contributing factor for the cellular response.
