Early Detection of Cymbidium Mosaic Virus and Odontoglossum Ringspot Virus in Tissue-Cultured Phalaenopsis Plantlets

Virus resistant Phalaenopsis breeding requires a lengthy time to make cultivar with qualification for obtaining practical virus resistance. We analyzed infection pattern and possibility for early detection of both CyrnMV and ORSV in tissue cultured Phalaenopsis plantlets to shorten the breeding time to selection of virus resistant Phalaenopsis. Both viruses could be detected by RT-PCR with specific primers for coat protein (CP) and by western blotting with respective CP specific antiserum in tissue cultured Phalaenopsis plantlets inoculated deliberately at the early stage (1-4 dpi) and we could monitor virus movement pattern from the site of initial inoculation to other parts of the leaf tissue by serial observation according to dpi. This study provides methods to confirm true virus resistance by mechanical inoculation for transgenic orchids against virus infection and can be adopted to raise selection efficiency. Therefore, it might be useful to shorten the breeding period with rapid detection ofvirus-resistant transgenic plantlets in vitro or check for virus free in tissue-cultured Phalaenopsis.
