The influence of different temperature regimes on in vitro multiplication of shoot cultures of Prunus glandulosa 'Alboplena' und Tilia cordata 'Wega' was investigated. The cultures were propagated on a modified MURA-SHIGE and SKOOG medium, at a PAR of 46 mumol m(-2) s(-1) 16 hours per day. Standard temperature was 22degreesC during lighting period and 20degreesC during dark period. The rise of daily mean temperature from 21,3degreesC to 24,3degreesC and 27,3 degreesC inhibited the shoot formation. If only the temperature of light or dark period was enhanced to 27degreesC, no inhibition of shoot formation was found. In some cases the shoot formation was even significantly higher. If the temperature of dark period was reduced from 20 degreesC to 17degreesC the shoot formation was inhibited. However, if the temperature of the lighting phase was reduced to 17degreesC no inhibition of shoot formation could be found. The results are discussed in respect of the concept of temperature gradient between dayight (DIF).
