In this postgenomic era there is a great demand in biomedical research for highly sensitive technologies that enable multiplexed detection and quantification of hundreds of low-abundance proteins and their modifications. For example, discovering effective biomarkers for improved prognosis/diagnosis of human diseases requires such technologies to rapidly verify and validate protein candidates. Traditionally, sensitive detection of low-abundance proteins (e.g., pg/ml to lowng/ml levels of proteins in human plasma/ serum) has primarily relied on immunoassays, such as ELISA, because of their specificity, sensitivity and throughput [1].
