A protocol for recovering nuclear halos from Sciaridae salivary gland polytene nuclei after the extraction of the nuclear proteins using lithium dioidosalicylate detergent was developed. This detergent removed almost 80% of the nuclear proteins, so that a DNA halo could be observed. The recovery of nuclear halos was only possible using hypotonic buffer free of spermine and spermidine.
