A protocol for recovering nuclear halos from Sciaridae salivary gland polytene nuclei after the extraction of the nuclear proteins using lithium dioidosalicylate detergent was developed. This detergent removed almost 80% of the nuclear proteins, so that a DNA halo could be observed. The recovery of nuclear halos was only possible using hypotonic buffer free of spermine and spermidine. The recovered nuclear halos from B. hygida salivary gland polytene nuclei contributed greatly to the study of the structure and function of these special organelles..
