Multiplication of Marine Unicellular Algae Nannochloropsis oculata Under Different Culture Media in Andaman Islands

Pipette method, washing method, serial dilution method, phototactic movement and agar plating method were carried out to isolate unicellular algae Nannochloropsis oculata. Among these methodologies, serial dilution was found suitable for its isolation. The pure strain obtained was then maintained in test tube in algal culture laboratoy under controlled optimum conditions. These cultures were used as inoculums for the media test in the stock culture experiments. The culture media such as Walnes, PM, SEAFDEC, TMRL, Suto, Miquels, Guillard f, f/2 and f/4 were used for the growth studies along with control.Out of the nine media tested, Walnes gave the highest population growth (18.625 lakhs cells/ml) on day 7 of culture. The other media which given better growth are in the order of Guillard f/2 (16.75 lakhs cells/ml on day 8), Guillard f/4 (16 05 lakhs cells/ml on day 8), Miquels (15.125 lakhs cells/ml on day 8), TMRL (13.75 lakhs cells/ml on day 8), SEAFDEC (12.35 lakhs cells/ ml on day 8), PM (11.55lakhs cells/ml on day 9), Guillard f (10.75 lakhs cells/ml on day 9), Suto (9.95 lakhs cells/ml on day 9 and control (7.27 lakhs cells/ml on day 9). The study also revealed that apart from Walnes medium, the media such as Guillard f/2 and f/4 are also suitable for its stock culture maintenance.
