DNA fingerprinting of pear genotypes to estimate genetic relationship using ISSR markers

Inter-simple sequence repeat (ISSR) markers were used for determination of the genetic relationships among 20 pear genotypes. The genus pyrus (pear) includes species of great diversity and these studies would also be desirable for the purpose of better management and conservation of the genetic resources and for planning the breeding strategies. In the reactions performed with 21 ISSR primers, 178 markers of pear cultivars were obtained. The size of fragments varied from 100 to 1100 bp. The greatestnumber of polymorphic products (11) was obtained in the reactions with primer ISSR-811. Degree of DNA polymorphism was estimated 96.62% (ISSR). The dendrogram derived from ISSR analysis grouped pear genotypes into two major cluster comprising of 18 (cluster A) and 2 (cluster B) genotypes which branched at similarity value of 0.43 and 0.69, respectively. Thus, the high level of detected polymorphism would impress their applicability in development of superior progenies, QTL mapping, molecular breeding, investigation of population genetic diversity, comparative mapping and the selection of the parents in near future.
