EFFECT OF DIFFERENT EXTENDERS, GLYCEROL LEVELS AND EQUILIBRATION TIMES ON DEEP-FREEZING OF BUCK SEMEN

A total of 32 ejaculates were collected from 4 Black Bengal bucks by electro-ejaculation method and were used to study the effect of 2 extenders ( Tris vs. Egg-yolk citrate ) and 2 level of glycerol ( 6.4% Vs. 9% ) inclusions in 4, 5 and 6 hours of equilibration times. The evaluation of semen quality was done before deep freezing and after thawing in respect of serum motility, % of live spermatozoa, % of morphologically abnormal sprem and % of detached/abnormal acrosome. Percentage of motile and livespermatozoa in both prefreezing and post-thawing conditions were found to be significantly higher in Tris extender with 6.4% glycerol level and 4 hours of equilibration time. It was also recorded that the above combination maintained significantly higher percentage of morphologically normal spermatozoa and intact acrosomes for both the prefreezing and post thawing conditions.
